Expression and function of DMT1 without IRE in C6 cells mediated by recombinant adenovirus

摘要: Divalent metal transporter 1 (DMT1) is a ferrous iron import protein. The improper expression of DMT1 is involved in neurodegenerative diseases. In the present study, we constructed a recombinant adenovirus containing the gene of DMT1 without the iron response element (DMT1-IRE) and investigated its expression and function in the C6 glioma cell line. The DMT1-IRE gene, obtained by RT-PCR, was cloned into the shuttle plasmid pAdTrack-CMV containing green fluorescent protein (GFP) reporter gene. Linearized plasmid pAdTrack-CMV-DMT1-IRE was subsequently co-transformed into ( ) BJ5183 cells along with an adenoviral backbone plasmid pAdEasy-1 after digestion with I. I-digested pAdEasy1-DMT1-IRE was then transfected into E1-transformed human embryonic kidney cells (HEK293 cells) , in which recombinant adenoviruses were generated within 7 to 10 days. The results demonstrated that we obtained the DMT1-IRE gene. pAdEasy1-DMT1-IRE yielded a large fragment, plus a smaller fragment of 4.5 kb after digestion with I. PCR confirmed pAdEasy1-DMT1-IRE contained gene DMT1-IRE, indicating the successful construction of recombinant adenovirus plasmid containing DMT1-IRE. GFP fluorescence further confirmed the generation of adenovirus. AdDMT1-IRE could efficiently infect C6 glioma cells. And cell viability decreased in Ad-DMT1-IRE infected cells after iron overload compared to the control. These results suggest that the over expressed DMT1-IRE can aggravate the iron induced cell death due to its iron influx function.

Loss of monocarboxylate transporter 1 aggravates white matter injury after experimental subarachnoid

摘要: Monocarboxylic acid transporter 1 (MCT1) maintains axonal function by transferring lactic acid from oligodendrocytes to axons. Subarachnoid hemorrhage (SAH) induces white matter injury, but the involvement of MCT1 is unclear. In this study, the SAH model of adult male Sprague-Dawley rats was used to explore the role of MCT1 in white matter injury after SAH. At 48 h after SAH, oligodendrocyte MCT1 was significantly reduced, and the exogenous overexpression of MCT1 significantly improved white matter integrity and long-term cognitive function. Motor training after SAH significantly increased the number of ITPR2+SOX10+ oligodendrocytes and upregulated the level of MCT1, which was positively correlated with the behavioral ability of rats. In addition, miR-29b and miR-124 levels were significantly increased in SAH rats compared with non-SAH rats. Further intervention experiments showed that miR-29b and miR-124 could negatively regulate the level of MCT1. This study confirmed that the loss of MCT1 may be one of the mechanisms of white matter damage after SAH and may be caused by the negative regulation of miR-29b and miR-124. MCT1 may be involved in the neurological improvement of rehabilitation training after SAH.

Cotransfecting norepinephrine transporter and vesicular monoamine transporter 2 genes for increased retention

Norepinephrine transporter (NET) transfection leads to significant uptake of iodine-131-labeled metaiodobenzylguanidine (131I-MIBG) in non-neuroendocrine tumors. However, the use of 131I-MIBG is limited by its short retention time in target cells. To prolong the retention of 131I-MIBG in target cells, we infected hepatocarcinoma (HepG2) cells with Lentivirus-encoding human NET and vesicular monoamine transporter 2 (VMAT2) genes to obtain NET-expressing, NET-VMAT2-coexpressing, and negative-control cell lines. We evaluated the uptake and efflux of 131I-MIBG both in vitro and in vivo in mice bearing transfected tumors. NET-expressing and NET-VMAT2-coexpressing cells respectively showed 2.24 and 2.22 times higher 131I-MIBG uptake than controls. Two hours after removal of 131I-MIBG-containing medium, 25.4% efflux was observed in NET-VMAT2-coexpressing cells and 38.6% in NET-expressing cells. In vivo experiments were performed in nude mice bearing transfected tumors; results revealed that NET-VMAT2-coexpressing tumors had longer 131I-MIBG retention time than NET-expressing tumors. Meanwhile, NET-VMAT2-coexpressing and NET-expressing tumors displayed 0.54% and 0.19%, respectively, of the injected dose per gram of tissue 24 h after 131I-MIBG administration. Cotransfection of HepG2 cells with NET and VMAT2resulted in increased 131I-MIBG uptake and retention. However, the degree of increase was insufficient to be therapeutically effective in target cells.

Comparison of the removal of monovalent and divalent cations in the microbial desalination cell

摘要: Microbial desalination cell (MDC) is a promising technology to desalinate water and generate electrical power simultaneously. The objectives of this study were to investigate the desalination performance of monovalent and divalent cations in the MDC, and discuss the effect of ion characteristics, ion concentrations, and electrical characteristics. Mixed salt solutions of NaCl, MgCl , KCl, and CaCl with the same concentration were used in the desalination chamber to study removal of cations. Results showed that in the mixed salt solutions, the electrodialysis desalination rates of cations were: Ca >Mg >Na >K . Higher ionic charges and smaller hydrated ionic radii resulted in higher desalination rates of the cations, in which the ionic charge was more important than the hydrated ionic radius. Mixed solutions of NaCl and MgCl with different concentrations were used in the desalination chamber to study the effect of ion concentrations. Results showed that when ion concentrations of Na were one-fifth to five times of Mg , ion concentration influenced the dialysis more profoundly than electrodialysis. With the current densities below a certain value, charge transfer efficiencies became very low and the dialysis was the main process responsible for the desalination. And the phosphate transfer from the anode chamber and potassium transfer from the cathode chamber could balance 1%–3% of the charge transfer in the MDC.

a novel K+ transporter gene in cotton

摘要: Potassium is an essential nutrient for plant growth and productivity of crops. K transporters are important for K uptake and transport in plants. However, information on the function of K transporters and K channels in cotton is limited. The KT/KUP/HAK protein family is essential for a variety of physiological processes in plants, including nutrient acquisition and regulation of development. This study, identified a K transporter gene, expressed in the roots of cotton ( ) cv. Liaomian17. The deduced transcript of is highly homologous to Cluster II of KUP/HAK/KT K transporters and is predicted to contain 11 transmembrane domains. has been localized to the plasma membrane, and its transcripts were detected in roots, stems, leaves and shoot apices of cotton seedlings. Consistently, b-glucuronidase (GUS) expression driven by the GhKT2 promoter could be detected in roots, mesophyll cells, and leaf veins in transgenic . In addition, the expression of was induced by low K stress in cotton roots and ::GUS-transgenic seedlings. The -overexpression lines plants were larger and showed greater K accumulation than the wild type (WT) regardless of K concentration supplied. The net K influx rate, measured by the noninvasive micro-test technique, in root meristem zone of -transgenic lines was significantly greater than that of WT. Taken together, this evidence indicates that GhKT2 may participate in K acquisition from low or high external K , as well as K transport and distribution in plants.

Identification of transporter proteins for PQQ-secretion pathways by transcriptomics and proteomics analysis

摘要: Pyrroloquinoline quinone (PQQ) plays a significant role as a redox cofactor in combination with dehydrogenases in bacteria. These dehydrogenases play key roles in the oxidation of important substrates for the biotechnology industry, such as vitamin C production. While biosynthesis of PQQ genes has been widely studied, PQQ-transport mechanisms remain unclear. Herein, we used both two-dimensional fluorescence-difference gel electrophoresis tandem mass spectrometry and RNA sequencing to investigate the effects of overexpression in an industrial strain of WSH-003. We have identified 73 differentially expressed proteins and 99 differentially expressed genes, a majority of which are related to oxidation-reduction and transport processes by gene ontology analysis. We also described several putative candidate effectors that responded to increased PQQ levels resulting from overexpression. Furthermore, quantitative PCR was used to verify five putative PQQ-transport genes among different PQQ producing strains, and the results showed that , and were upregulated in all conditions. Then the three genes were over-expressed in WSH-003 and PQQ production were detected. The results showed that extracellular PQQ of B932_1930 (a transporter) and B932_2186 (an ABC transporter permease) overexpression strains were enhanced by 1.77-fold and 1.67-fold, respectively. The results suggest that the proteins encoded by PqqB, B932_1930 and B932_2186 might enhance the PQQ secretion process.

摘要: 提出一款应用于第五代金属边框智能手机的高隔离度两天线模块。基于一个蚀刻在地板上的T形缝隙,通过合理地设计一个耦合馈电环天线和一个耦合馈电缝隙天线,实现了一款紧凑的两天线模块。尽管耦合馈电环天线和耦合馈电缝隙天线共口径,两者之间的隔离度高于30 dB。随后,通过集成4个两天线模块,实现了一款8×8多入多出系统。该系统实测互耦和包络相关系数分别小于−18.5 dB和0.02。

摘要: 随着处理器设计转向使用专门的异构多核以避免功耗墙的影响,软件开发人员发现很难处理这些处理器系统的复杂性。以Matrix-3000为代表的新型处理器具有复杂的内存层次结构和处理器组织,是为下一代E级超级计算机设计的高性能处理器。本文分享了我们为Matrix-3000开发的并行编程模型及其支持编译器和库的经验。为了帮助软件开发,我们从头开始开发了一个针对Matrix-3000的软件栈,包括一个低层次的编程接口和一个高层次的OpenCL编译器。该低层次编程模型为使用Matrix-3000的裸金属加速器提供了原生编程支持,而高层次模型允许程序员使用OpenCL并行编程标准。我们详细介绍了该软件栈的设计选择,并强调了从开发系统软件中学到的经验教训,以实现裸金属加速器的高效程序编写和性能解锁。我们的编程模型已经被部署到一个E级原型系统的生产环境中。

A ternary mechanism for the facilitated transfer of metal ions onto metal–organic frameworks: implications

摘要: Although metal–organic frameworks offer a new platform for developing versatile sorption materials, yet coordinating the functionality, structure and component of these materials remains a great challenge. It depends on a comprehensive knowledge of a “real sorption mechanism”. Herein, a ternary mechanism for U(VI) uptake in metal–organic frameworks was reported. Analogous MIL-100s (Al, Fe, Cr) were prepared and studied for their ability to sequestrate U(VI) from aqueous solutions. As a result, MIL-100(Al) performed the best among the tested materials, and MIL-100(Cr) performed the worst. The nuclear magnetic resonance technique combined with energy-dispersive X-ray spectroscopy and zeta potential measurement reveal that U(VI) uptake in the three metal–organic frameworks involves different mechanisms. Specifically, hydrated uranyl ions form outer-sphere complexes in the surface of MIL-100s (Al, Fe) by exchanging with hydrogen ions of terminal hydroxyl groups (Al-OH2, Fe-OH2), and/or, hydrated uranyl ions are bound directly to Al(III) center in MIL-100(Al) through a strong inner-sphere coordination. For MIL-100(Cr), however, the U(VI) uptake is attributed to electrostatic attraction. Besides, the sorption mechanism is also pH and ionic strength dependent. The present study suggests that changing metal center of metal–organic frameworks and sorption conditions alters sorption mechanism, which helps to construct effective metal–organic frameworks-based sorbents for water purification.

Brazing of ceramic-to-ceramic and ceramic-to-metal joints in air

摘要: Reactive air brazing (RAB) is an emerging technology for the production of ceramic-to-ceramic and ceramic-to-metal joints. In this study, RAB was investigated with respect to the potential applications for solid oxide fuel cells (SOFCs) as one example of use. It was found that alumina could be well brazed by RAB with AgCu and AgCuTi brazes. Both braze composition and brazing temperature influenced significantly the wetting behavior and their mechanism of wetting. AgCu and AgCuTi braze alloys could also be used to produce brazed joints with the SOFC materials ceramic yttria stabilized zirconia and steel X1CrTiLa22. However, CuO reacts with the steel, forming a brittle oxide layer on the steel surface, which is undesirable for SOFC applications. The first trials with Ag0.5Al showed a promising solution.

Optimizing iodine capture performance by metal–organic framework containing with bipyridine units

摘要: Radioactive iodine exhibits medical values in radiology, but its excessive emissions can cause environmental pollution. Thus, the capture of radioiodine poses significant engineering for the environment and medical radiology. The adsorptive capture of radioactive iodine by metal–organic frameworks (MOFs) has risen to prominence. In this work, a Th-based MOF (denoted as Th-BPYDC) was structurally designed and synthesized, consisting of [Th63-O)43-OH)4(H2O)6]12+ clusters, abundant bipyridine units, and large cavities that allowed guest molecules diffusion and transmission. Th-BPYDC exhibited the uptake capacities of 2.23 g·g−1 and 312.18 mg·g−1 towards I2 vapor and I2 dissolved in cyclohexane, respectively, surpassing its corresponding analogue Th-UiO-67. The bipyridine units boosted the adsorption performance, and Th-BPYDC showed good reusability with high stability. Our work thus opened a new way for the synthesis of MOFs to capture radioactive iodine.

Selective capture and separation of xenon and krypton using metal organic frameworks: a review

摘要: Xenon and krypton are widespread useful noble gases in commercial lighting, lasers, electronics, and medical industry. At the same time, radioactive noble gases may proliferate from used nuclear fuel and diffuse in open atmospheres. Metal organic frameworks as hotspot porous materials for gases uptake and separation are considered to be potential solutions. In this review, we comprehensively summarized recent researches on metal organic frameworks for selective capture and separation of xenon and krypton. Particularly, we followed the aspects of different optimal design strategies, including optimal pore/cage size and geometry, open metal sites, ions (anions and cations), and polar functional groups for enhancing the xenon adsorption and separation performances. Meanwhile, a comparison of each strategy and the mechanisms of xenon/krypton separation were pointed out. The separation of krypton from gases mixtures by dual-bed systems was further discussed. Finally, some existing challenges and opportunities for possible real applications were proclaimed.

Liquid metal thermal hydraulics R&D at European scale: achievements and prospects

摘要: A significant role for a future nuclear carbon-free energy production is attributed to fast reactors, mostly employing a liquid metal as a coolant. This paper summarizes the efforts that have been undertaken in collaborative projects sponsored by the European Commission in the past 20 years in the fields of liquid-metal heat transfer modeling, fuel assembly and core thermal hydraulics, pool and system thermal hydraulics, and establishment of best practice guidelines and verification, validation, and uncertainty quantification (UQ). The achievements in these fields will be presented along with the prospects on topics which will be studied collaboratively in Europe in the years to come. These prospects include further development of heat transfer models for applied computational fluid dynamics (CFD), further analysis of the consequences of fuel assembly blockages on coolant flow and temperature, analysis of the thermal hydraulic effects in deformed fuel assemblies, extended validation of three-dimensional pool thermal hydraulic CFD models, and further development and validation of multi-scale system thermal hydraulic methods.
